SSB-Seq

SSB-Seq

Map DNA single-strand breaks (SSB-Seq) in vivo. In this method the genomic DNA is isolated and subjected to nick-translation by DNA polymerase I in the presence of deoxynucleotide triphosphates including digoxigenin labeled dUTP and a small amount of dideoxynucleotides. The DNA was sheared and the labeled fragments were immunoprecipitated with anti-digoxigenin antibody and purified.

Pros:
  • Simple protocol
Cons:
  • The locations of the breaks cannot be determined accurately
  • Not adopted by the community
