
Cap analysis gene expression sequencing (CAGE) is a protocol that can measure RNA expression and map transcription start sites (TSS) in promoters. In this method, RNA is first reverse transcribed using a random primer. RNA cap and 3’-ends are biotinylated. Non-hybridized single-stranded RNAs are digested with RNase, leaving 5’ complete cDNAs which are captured using streptavidin beads. The cDNA is then processed for sequencing.
